Gladney entered her plea before U.S. Magistrate Jerry A. Davis in the Aberdeen federal courthouse.
A July 6 trial date was set before U.S. District Judge Sharion Aycock in Aberdeen.
Gladney was indicted by a federal grand jury April 23, but the formal accusation was not unsealed until earlier this week. She posted a $10,000 bond after an initial court appearance Tuesday.
She is accused of illegally using more than $5,000 in federal AmeriCorps literacy program funds to pay some of her day care workers, who were not providing AmeriCorps services.
If convicted, she faces up to 55 years in prison and $2.25 million in fines.
